MendelWorks offers three approaches to genotype your mice:
Standard PCR – We verify the assay information and lab protocols you provide to us. Then, your assay is replicated and optimized for use in our lab. Once we have made the assay robust and reproducible, we are ready to receive regular genotyping orders from you. Gel images of the PCR data are provided with the genotyping results report.
Real-time PCR – We can develop a real-time PCR assay to screen your mutant. Real-time PCR assays have many advantages over standard gel-based PCR assays, including high specificity and faster turnaround times. Design of real-time assays requires clear sequence information of the mutant being tested. If needed, we will sequence the mutant region of genome to facilitate the validation process. Our Genotyping Consultants can discuss the advantages and requirements of real-time assays with you to help you make an informed decision.
Quantitative PCR – If it is important to know the copy number of a transgene, we can provide you with highly accurate, quantitative information. Two types of service are offered:
Transgene copy number per genome, relative to a standard curve, or
Zygosity testing for lineages derived from known founders. This method provides a high level of confidence in copy number of tested animals relative to known heterozygotes for that line. All quantitative assays are run in triplicate in two separate experiments.
